Is it possible to crop video in anything but rectangular shapes?

And on a side note, is it possible to do the crop operation in the API?

That would be the new "luma key" coming up in vMix 13:

And download the newest (but unstable) version of vMix here, which includes this feature

As for the second question, no, it does not as far as I know.
