I'm bringing in a source from Desktop Capture with audio...I want to use that audio as my main Output audio regardless of which source I'm playing. How do I configure that? Is there a video on setting up Output audio somewhere that I haven't found yet?

I keep doing it wrong! Echoes or mute when I change source etc. And I don't want to hear it on the vMix machine...I just want it output. Do I have to just mute the vMix machine speakers? I know there's mapping to this...

So if I turn off auto mix on the NDI input and the headphone knob down... hopefully in doing that right.

The double arrow icon of each Input with Audio in the vMix Audio Mixer is for "Audio follows Video", meaning the Audio for that Input will be "ON" when the Input is active in Program and "OFF" when not.
To have "Audio Always ON" for an Input, you need to deactivate that icon and activate the speaker icon so that it is always "ON".
To insure audio is always "OFF" for an Input, you are best to lower it's fader to 0% in vMix's Audio Mixer.
