At the moment the multiview layout that is selectable as a source from the fullscreen dropdown is very strict. It only allows a few preset layouts and nothing more. I know I can set fullscreen to an input and make something more custom there but I need access to the preview and output screens which inputs currently do not have.

Output screen is the largest screen with 4 on the bottom (game, players, casters) and a chat window on the side

You could use vMix Output 1 NDI as program and Output 3 NDI as Preview, then add them to vMix using NDI source. This way you can build your own multiview with preview/program.
