I'm trying to set up a simple shot with a Zoom guest being one of the inputs, but the input I create for the Desktop capture is not activating audio. I can't turn it on or off and no audio for "Desktop Capture" appears in the mixer.

I've done this multiple times with other software, and I know there is a way, so any help would be greatly appreciated.

some time I need to stream games, but it seems that vmix can not set the game sound as the audio input?

Probably more than one way to do it, but go to Add Input, then Audio Input to add the audio from the screen capture.

Well, there is no obvious "Desktop Capture" source as an audio input. Just my normal microphones, virtual microphones, etc.

Even if I add any audio source to my Desktop Capture input, it won't share the actual audio of the desktop, or even let me click the "Audio" button under the input.

Or you can use Virtual Audio Cables and bring in your Zoom guest on the same PC as vMix. Have a look here:
