Does the vMix gain (input options: general/audio settings) has another behaviour then the gain input of my soundcard?
BTW can I decrease the gain of my soundcard to avoid clipping and increase vMix gain to compensate the volume?
When the audiolevel of my soundcard >0dB I've got a loud distorsion. I guess the vMix gain is a post-soundcard gain, can it also produce clipping when set to high?

This is one of the downsides to Windows audio devices. Any Windows audio devices goes through what is called the KMixer. It's hard to tell exactly what is happening to your audio when it goes through it but it could get resampled or the gain structure could change depending on the audio device that is being used. That's an unfortunate issue when routing audio out and back into an audio device on your computer. When using a digital signal like RCA S/PDIF or Toslink connection, some of these issues can be minimized. The other downside is the amount of A/D and D/A conversion that happens at each step in the chain. This is where gain structure can get really messed up and where correct audio processing can be crucial especially when working between multiple audio devices in a chain.
What audio device/card are you using? Are you doing any signal processing along the way? EQ, Compression, Limiting, Conversion?

With regards to gain, this is how digital audio works. (And not specifically limited to windows)
If an audio source in vMix shows up on the meter close to 0db, then there is no more gain possible as you are already reaching the maximum volume possible for a given bit rate.
So gain in vMix can only compensate for a low source volume, but can't increase overall volumes beyond 0db. So any further gain would need to be done by the output audio device.

I've got a Osprey 440 audio/video grabbercard. The Analog prescale is set to -5dB and software boost is 0dB. So I guess because I've got some headroom, the gain in vMix has no advantage for me.
I can't prevent some clipping because with a lower setting my stream volume is to low and people have to turn up their volume.
