A real time saver and usability improvement would be to bring the PTZ controls out into the main UI. Having to dig into Settings menus to get at the PTZ controls is a little clunky.

There are quite a few different ways this could be achieve so I'll suggest a few possible ways to get the conversation started.

METHOD 1
The control buttons associated with each camera input would have a PTZ control selector button. Click on the button will send any PTZ commands out to the selected camera. Only one input can be selected at a time.

Just to the left of the Audio section would be a PTZ control panel like vMix already has inside the Settings for each input. Pressing those controls send the movement commands to the selected camera.

I believe this is similar to what Wirecast is going to be doing in the upcoming v9.

METHOD 2
Have two sets of PTZ control buttons under the Preview and Output windows. This will allow easy control of the live shot and gives the vMix operator the opportunity to get the shot framed correctly in preview.

Buttons: Left, up, down, right, focus + and -, exposure + and -, etc.

METHOD 3
Put PTZ controls under and to the right side of each camera input.

Handling preset positions would be an added benefit if that could be exposed as well.

As vMix already support PTZ controls, I suspect this is only a matter of making some UI changes and hooking into the camera control functions already present in vMix.

+1, I think it could be nice to have seperate screens for seperate tasks. vMix has now become so powerful, that multiple people could be controling a single machine. Apart from PTZ control, where an rcp operator could take a seperate screen, one for a color corrector/shader would be useful too.
